It has been known for many years that the subcellular distribution of proteins throughout eukaryotic cells is highly ordered. This means that proteins have to be transported to their correct destinations and left there. Understanding the complex mechanisms by which this occurs has provided a major challenge for modern biology. Recent progress in the subject has relied heavily on finding ways to measure protein transport and targeting. This work brings together contributions from active researchers in the field, who provide step-by-step protocols for studying the processes involved, using both intact cells and broken cell preparations. Cell biological, biochemical and molecular biological methods are all addressed in detail, and the chapters cover the well-established techniques, as well as a number of new methodologies which have potentially very wide applications. This text aims to enable scientists from varied backgrounds to get to grips with unravelling the mechanisms which control cellular protein targeting.
