Le but de l'étude était de préparer et d'évaluer le gel flottant in situ de méthotrexate pour augmenter le temps de séjour gastrique résultant en une libération prolongée du médicament dans le tractus gastro-intestinal. Le méthotrexate est un médicament anticancéreux. Il montre l'effet secondaire indésirable en raison de leur libération rapide dans le tractus gastro-intestinal (GIT). Le gel flottant in situ a été préparé en utilisant des polymères formant du gel in situ comme l'alginate de sodium et la pectine. La compatibilité médicament-excipients a été réalisée par étude FT-IR. Une conception factorielle complète à 3 niveaux 2 facteurs a été appliquée pour étudier l'effet combiné de la concentration d'alginate de sodium (X1) et de la concentration de carbonate de calcium (X2). Les gels flottants in situ ont été évalués pour le temps de latence flottant, le % de libération de médicament à 1 heure et le % de libération de médicament à 12 heures. En plus de ce qui précède, les gels flottants in situ ont également été évalués pour le pH, la viscosité, la capacité gélifiante, le temps de flottaison, la teneur en médicaments. Toutes les formulations ont donné un résultat satisfaisant en termes de temps de latence flottante, % de libération de médicament à 1 heure et 12 heures. La libération du médicament dépend de la concentration du polymère et du carbonate de calcium utilisés dans la formulation.
