Nach den Staatsexamina und der Promotion war Inez De Florio-Hansen zunächst mehrere Jahre als Fremdsprachenlehrerin tätig. Nach der Habilitation lehrte sie von 1996 bis zur Emeritierung als Universitätsprofessorin Fremdsprachenforschung und Interkulturelle Kommunikation an der Universität Kassel. Seit der Jahrtausendwende haben sich die Forschungsschwerpunkte von Inez De Florio-Hansen deutlich erweitert, nämlich hin zu Unterricht auf empirisch-experimenteller Grundlage sowie zur Berücksichtigung von erziehungswissenschaftlichen Erkenntnissen als Ergänzung zu fremdsprachendidaktischen Überlegungen. Einen weiteren Schwerpunkt bildet die Digitalisierung in Schule und Unterricht.
