Synopsis

An experiment was conducted to evaluate the performance of five wheat varieties viz. Gourav, Shatabdhi, Sonalika, Prodip and Barkat in respect of mature embryo culture and different concentrations of growth regulators ; 2, 4-D, kinetin, BAP, IAA and AgNO3. Mature embryos from five varieties were placed in MS medium supplemented with 2, 4-D (0, 2.5, 3.0, 3.5, 4.0 and 4.5 mg/L) and kinetin (0.5 mg/L) for callus induction on BAP (0, 1.0, 1.5, 2.0, 2.5 and 3.0 mg/L), IAA (1.5 mg/L) and AgNO3 (2.0 mg/L) for plant regeneration. Callus growth was measured by size using transparent scale. There were significant differences among the varieties in callus size. Barkat showed the best callus size (5.154 mm) on MS medium supplemented with 3.5 mg/L 2, 4-D and 0.5 mg/L Kinetin. In addition, phytohormone had significant effect in callus induction. In fact the proper dose of hormone used, the highest callus (3.650) found from MS+ 3.0 mg/L 2, 4-D +0.5 mg/L Kinetin. Among the varieties, Shatabdhi showed the best performance for shoot regeneration.Variety and hormone concentration had significant effect in shoot regeneration. Sonalika and Shatabdhi showed highest performance for root initiation.
