Synopsis
Les tumeurs malignes représentent l'une des maladies humaines les plus courantes et malgré les progrès révolutionnaires en sciences thérapeutiques, le pronostic clinique du cancer reste relativement faible. Cela laisse donc suffisamment de place pour le développement de nouvelles pistes thérapeutiques ou de stratégies pour l'amélioration des médicaments actuellement utilisés. Parmi les cibles actuelles de la chimiothérapie, avec l'ADN, les microtubules ont un rôle de premier plan à jouer. Étant donné que les structures contenant de la tubuline sont cruciales dans un certain nombre de fonctions cellulaires, les microtubules ont représenté une bonne cible pour la chimiothérapie. Un certain nombre de médicaments, principalement d'origine naturelle, y compris la vinblastine, la colchicine et le paclitaxel ont émergé pour cibler les microtubules sur divers sites de liaison. De même, les composés qui se lient aux cibles basées sur l'ADN comme Top I et Top II (par ex. les analogues dérivés de la camptothécine et de la podophyllotoxine) représentent également les principaux résultats thérapeutiques pour contourner les tumeurs malignes.
