Nadolol is a BCS Class III drug having low permeability and high solubility. It has low oral bioavailability. Formulation of Nadolol into wafers will help in improving the delivery of the drug. The design of the formulation started with the factorial design to optimize the independent variables like the amount of PEG, SLS and SSA. Formulation batches of wafers were prepared in accordance with Box BehnKen Design and evaluation tests were performed. It was observed that the independent parameters concentration of PEG, SLS and SSA show impact on disintegration time, folding endurance and thickness of the wafers. The data obtained in the factorial experiments was used for plotting response-surface and contour plots and data was analyzed by ANOVA. Optimized formulae were given by the software. The optimized formulations were prepared and evaluated. The formulation having the lowest % relative error of the predicted values given by the software and the experimental values was considered the best. OF1 was found to show better results. The optimized formulation of nadolol loaded wafers was then subjected to in vitro drug release studies.
