Synopsis

Des perfusions de cellules T et de cellules NK (Natural Killer) provenant du sang périphérique sont effectuées pour traiter les tumeurs malignes. Les lymphocytes propagés ex vivo, en normoxie (20 % d'O2), sont perfusés par voie intraveineuse et doivent survivre à l'hypoxie associée à la circulation veineuse, à la moelle osseuse (5 % d'O2) et au microenvironnement tumoral (1 % d'O2). L'objectif principal de cette étude était de déterminer la capacité de prolifération des cellules T et NK humaines en normoxie (20 % d'O2) par rapport à l'hypoxie (1 % d'O2), pendant 28 jours, en utilisant une cellule présentatrice d'antigène artificielle (aAPC) pour une propagation de niveau clinique.
