Torr. from the absolute nuclear DNA content using leaves and a flow cytometry-based method. Leaves were processed with a simple mincing technique in a buffer solution to obtain cell nuclei in suspension, which were stained with propidium iodide and then analyzed with an Attune� acoustic focusing cytometer (Applied Biosystems) of the Flow Cytometry area of the National Laboratory for Agri-Food and Forestry Research and Service (LANISAF). For this purpose, leaf tissue nuclei from Zea mays L. were used as reference standard with DNA value 2C=5.67 pg.
