Our the- pies for tumors have mostly been based on classical chemotherapy and antiproliferative treatments. More recently, directed therapies against a causative oncogene have led to prominent reduction in tumor rates for some cancers. For instance, chronic myelogenous leukemia that is due to a BCR¿ABL translocation can be targeted with Gleevec. Despite these advances in individual tumors, a number of patients are treated for their primary tumors and ultimately relapse. Relapsing tumors can be due to resistance to chemotherapy or to antioncogene drugs. Another hypothesis is that heterogeneity in the tumor leads to an inability of classical chemotherapy to completely eradicate all cells of the tumor. This concept of heterogeneity has led some investigators to propose a cancer stem cell model. As patients are treated with chemotherapy, most of the dividing cells are killed, but this leaves a small subset of cells that have the ability to remake the entire tumor. These are cancer stem cells. They possess the signals of self-renewal and yet can also differentiate. If one could understand more about the cells that remain after classical chemotherapy or the cells that can remake the tumor among the heterogeneous population, this information would have a huge impact on our treatment of cancer.Humana Press in Springer Science + Business Media, Heidelberger Platz 3, 14197 Berlin 300 pp. Englisch.

The target audience includes clinical or basic scientists who look to apply proteomics to the neurosciences. Often researchers hear of proteomics without an adequate explanation of the methodology and inherent limitations. This volume conveys where proteomic methodology is in its application to CNS research and what results can be expected. We also address clinical translation of neuroproteomics, specifically in the area of biomarker research. The inception of neuroproteomics capitalized on rapid progress in large-molecule mass spectrometry over the last decade. Two seminal advances have spurred research ¿ development of reliable polypeptide ionization processes and bioinformatics to rapidly process tandem mass spectra for peptide identification and quantification. What has followed is the exponential application of mass spectrometry to proteome characteri- tion across biological and biomedical disciplines. Arguably, the most elaborate proteomic implementation is in studying the CNS, the most enigmatic and complex animal system. Neuroscience is characterized by grandiose questions ¿ what is consciousness, how does thought or memory work. Neuroproteomics researchers, however, have pri- rily involved themselves dysfunction, based on a pressing need (and invariably funding), in answering questions on CNS dysfunction, based on a pressing need (and invariably funding), and because such questions hold more accessible answers. Dysfunction is readily contrasted against normal function and presumably produces a lasting differential protein signature.Humana Press in Springer Science + Business Media, Heidelberger Platz 3, 14197 Berlin 336 pp. Englisch.

In Life-Span Extension: Single-Cell Organisms to Man, the editors bring together a range of illuminating perspectives from researchers investigating the aging process in a variety of species. This novel work addresses the aging process in species ranging from yeast to man and, among other subjects, features detailed discussions of the naked mole-rat, an exceptionally long-lived rodent; the relationship between dietary factors/food restriction and aging; and an evolutionary view of the human aging process.Single mutations that extend life span have been identified in yeast, worms, flies, and mice, whereas studies in humans have identified potentially important markers for successful aging. At the same time, it has been discovered that the genes and pathways identified in these studies involve a surprisingly small set of conserved functions, most of which have been the focus of aging research for some time. For example, the mTOR pathway, a regulator of translation and protein synthesis, has been identified as a common longevity pathway in yeast and Caenorhabditis elegans. In mammals, this pathway intersects with neuroendocrine pathways and with the insulin/insulin-like growth factor pathways, which have been identified as major modulators of life span and aging in both invertebrates and mice.Novel, emerging technologies and the increasingly wide variety of systems that are now used to study aging and the mechanisms of aging provide enormous opportunities for the identification of common pathways that modulate longevity. It is these common pathways that are the focus of this important volume.Humana Press in Springer Science + Business Media, Heidelberger Platz 3, 14197 Berlin 216 pp. Englisch.

In Chromatin Immunoprecipitation Assays: Methods and Protocols, researchers deeply involved in the development and improvement of the field provide cutting-edge protocols devoted to the most recent progress in ChIP and related subjects. The thorough chapters involve topics such as the characterization of ChIP antibodies, ChIP methods for small cell numbers, fast ChIP protocols, assays adapted to various species and cell types, as well as several strategies for the analysis of genome-wide datasets, while also extending a bit beyond ChIP assays to cover immunoprecipitation-based DNA methylation analyses, determination of spatial chromatin organization of large genomic regions, and RNA immunoprecipitation. Written in the highly successful Methods in Molecular Biology¿ series format, chapters contain brief introductions to their respective subjects, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and notes on troubleshooting and avoiding known pitfalls.Authoritative and easy-to-use, Chromatin Immunoprecipitation Assays: Methods and Protocols is a timely guide for all those who wish to further the development of this powerful tool and continue delving into the incredible complexity of the cell¿s biology.Humana Press in Springer Science + Business Media, Heidelberger Platz 3, 14197 Berlin 280 pp. Englisch.

Biology of Stem Cells and the Molecular Basis of the Stem State also introduces the reader to the commonly accepted notions regarding stem cell biology, with an emphasis on an alternative view of stemness, i.e. the stem state. In keeping with the popularity of this topic, Biology of Stem Cells and the Molecular Basis of the Stem State addresses the major controversies and points of dispute, among researchers in the stem cell field. Overall, Biology of Stem Cells and the Molecular Basis of the Stem State presents a well-rounded dialogue about stem cells as it not only concentrates upon the biological elements of stem cell, but also addresses the controversy and hype currently enveloping this popular subject.Humana Press in Springer Science + Business Media, Heidelberger Platz 3, 14197 Berlin 304 pp. Englisch.
