Druck auf Anfrage Neuware - Printed after ordering - In this project we bred together PDGF-B retention motif knockout (PDGF- Bret) mice and PDGF-C knockout mice for the first time to investigate the effect of combined PDGF-B and PDGF-C deficiency on mouse development and brain vasculature.PDGF-Bret/ret;P…DGF-C-/- double knockout mice were born dead and alive, but none survived weaning. Though severe oedema and haemorrhage was observed, they did not show any specific macroscopic malformation. The brain vasculature of new born (P0) PDGF-Bret/ret;PDGF-C-/- double knockout mice seemed to present a lower density and a more distorted architecture compared to PDGF-Bret/ret;PDGF-C+/-,PDGF-Bret/ret mice, but no significant changes could be shown.Comparing the brain vasculature of adult PDGF-Bret/ret;PDGF-C+/- and PDGF-Bret/ret mice we could not show significant differences of vascular density, but we were able to detect significant higher perimeter and length of the vessels in some parts of the brain. Looking at mural cells we could not show significant changes in pericyte architecture, but our observations suggest a thicker vSMC coverage of big vessels in PDGF-Bret/ret;PDGF-C+/- mice, an effect already described in PDGF-C-/-mice (Fredriksson et al., 2012).In conclusion, our data suggests that combined PDGF-B and PDGF-C deficiency in mice allows a normal macroscopic embryonic development, but limits surviving after birth presumably also due to severe malfunctions of the angiogenic system. Our histopathologic analysis of the brain vasculature underlines the described effect of PDGF-B and PDGF-C on the neurovascular unit, but suggests that the interaction and redundancy of these two PDGFs is not sufficiently distinct for a partial deficiency leading to extreme phenotypic changes.

Nevertheless, the recent progress in genetics will allow us to contem plate, for the first time, the integration of observation into a coherent view of brain development. Clearly, this may be a major challenge for the next century, and arguably is the most important task of contemporary develop mental biology. The purpose of the present book is to provide an overview that syn thesizes up-to-date information on selected aspects of mouse brain devel opment. Given the format, it was not possible to cover all aspects of brain development, and many important subjects are missing. The selected themes are, to a certain extent, subjective and reflect the interests of the contributing authors. Examples of major themes that are not covered are peripheral nervous system development, including myelination, the development of the hippocampus and several other CNS structures, as well as the developmental function of some important morphoregulatory molecules.

Although this interesting mutation has been with us for half a century, it is fair to say that, in spite of a wave of en…thusiasm in the late sixties and early seventies, generated primarily by Sidman, Caviness and colleagues, studies of reeler mice fell pro gressively out of fashion during the next two decades. All that changed almost overnight when the cloning of the reeler gene, dubbed reelin, was reported in Tom Curran's laboratory in 1995. The fact that the same gene was identified at the same time independently by two other groups using positional cloning sug gested strongly that reelin was the right candidate. Although the key experiments of transgenic rescue have not been made (and perhaps will never be), the equation 'reeler is reelin' has been established beyond reasonable doubt, as alterations of the reelin gene and/or its expression have been found in at least five alleles of reeler and in the mutation Shaking Rat Kawasaki (SRK), an ortholog of reeler.

There is no significant difference in the relative neuronal and astrocyte frequency between the aggregation and ESC chimeras. Both chimera types do not differ in the number of neurons and supporting cells differentiated from PG donor cells. The potential of PG stem cells to form functional dopaminergic neurons was analyzed comparatively in the two chimeric types. Dopaminergic neurons derived from PG donor cells were detected in both chimeric types. However, the expression of many genes also involved in organogenesis correlates with the expressio…n of genes of Wnt and Bmp families and their effectors. In particular, one of such genes is Hr (Hairless) gene. Mutation in the Hr gene in mice leads to the development of a characteristic phenotype expressed in complete baldness by three weeks of age. Also, adult mice carrying the mutant allele of the Hr gene in the homozygote show anatomical defects in the thymus, inner ear, brain and intestine, organs whose tissues express the normal Hr gene in heterozygotes and wild-type animals. The formation of these organs occurs during embryogenesis and is controlled by poorly understood molecular mechanisms. Inductive interactions between epithelial and mesenchymal components of embryonic setting play an important role in the regulation of their development.The aim of this work is to find out whether the normal allele of Hr gene affects the functioning of WNT- and BMP-dependent gene networks in the embryogenesis of epithelio-mesenchymal organs in mouse. 64 pp. Englisch.

However, the last few years have wit nessed a quantum leap in our knowledge, due to technological improve ments, particularly in mole…cular genetics. Despite this progress, the available body of data remains mostly phenomenological and reveals very little about the grammar that organizes the molecular dictionary to articulate a pheno type. Nevertheless, the recent progress in genetics will allow us to contem plate, for the first time, the integration of observation into a coherent view of brain development. Clearly, this may be a major challenge for the next century, and arguably is the most important task of contemporary develop mental biology. The purpose of the present book is to provide an overview that syn thesizes up-to-date information on selected aspects of mouse brain devel opment. Given the format, it was not possible to cover all aspects of brain development, and many important subjects are missing. The selected themes are, to a certain extent, subjective and reflect the interests of the contributing authors. Examples of major themes that are not covered are peripheral nervous system development, including myelination, the development of the hippocampus and several other CNS structures, as well as the developmental function of some important morphoregulatory molecules. 356 pp. Englisch.
