We hypothesized thatgenetic variants in the estrogen receptor alpha(ERalpha) gene may cause variable sensitivity to thehormone and influence an individual s risk ofcardiovascular disease, along with otherestrogen-sensitive phenotypes. This work explores therole of ERalpha in the mediation of theatheroprotective effects of estrogen and identifiesregulatory regions in the ERalpha gene important incardiovascular tissue. It identifies novel geneticvariants in these regions and examines their effecton expression of the receptor in vitro, and theirassociation with estrogen sensitive phenotypes indifferent study populations. The identification ofgenetic variations associated with altered estrogenresponse is of potential public health importance.
